Genetic analysis of resistance to late blight in a diploid potato population derived from Solanum verrucosum

The mexican wild potato species Solanum verrucosum is considered as a potentially valuable source of resistance to Phytophthora infestans. The progeny (VPT) from a cross between a highly resistant hybrid S. verrucosum x S. phureja (VP45) with the dihaploid PS3 clone of S. tuberosum was tested for the presence of race-specific R genes. Both, the detached leaf assay and inoculation on the whole plant indicated no presence of a known major gene. Field evaluation at two locations of the VPT population revealed the segregation of LB resistance with continuous variation, indicative of a quantitative trait. Good correlations (r = 0.82) were observed between both data sets. A total of 165 clones from this family were genotyped with 100 AFLP primer combinations. A genetic linkage analysis was performed for both parents. For the female parent, 386 markers grouped 12 linkage groups; nine of them were anchored to known potato chromosomes with microsatellite mapped markers. Linkage analysis of the male parent with 252 markers resulted in 12 linkage groups, of whiwere ass potato chromosome revealed by the non-parametric Kruskal–Wallis test and interval mapping analysis revealed similar results. QTLs effects were observed on two unassigned linkage groups from the female parent only for one field evaluation. Two major and reproducible QTLs were detected on paternal chromosomes 2 and 3, explaining respectively 19.6 and 26%, of the phenotypic variation. The QTL on chromosome 3 had its highest LOD score (10.36) and is tightly associated with the microsatellite marker STM1053. This marker could be useful for late blight resistance breeding.
