Antidepressant-like effect of Omega-3 and extracts from Ginseng and Valeriana in combination in mice

Abstract: Objective Previous research had shown that Ginseng total saponins, Omega-3 and Valeriana had antidepressant-like effect. Here we investigated the antidepressant-like effects of a complex composing of those three ingredients in mice forced swimming test and tail suspension test. Method C57BL/6 mice were randomly divided into 5 groups, which received oral administration of imipramine (30mg/kg), low dose complex (225mg/kg), medium dose complex (450mg/kg), high dose complex (900mg/kg) and saline respectively. Animal continuously received treatment at 24 hour, 1 hour, 0.5 hour, before behavioral test including tail suspension test, forced swimming test and locomotor activity test. Result It was observed that complex at dose of 225, 450 and 900 mg /kg significantly reduced the immobility time in TST [F(4,25)=8.866,P<0.01 ] and FST [F(4,25)=6.982, P<0.01 ] in mice compared with the saline, uniformly with the imipramine. Interesting, the complex at each dose didn’t affect the locomotor activity in mice, and imipramine significantly reduced the locomotor in mice on the contrary. It indicated that antidepressant effect of complex seemed unlikely to be due to an increase in locomotor activity of mice. Conclusion The results of the observation suggest that the complex composing of Ginsengtotal saponins, Omega-3 and Valeriana have antidepressant effect in mice, meanwhile don’t to decrease locomotor activity of mice comparison with imipramine.
