BRIEF RESUME OF THE INTENDED WORK

6.1 NEED FOR THE STUDY

Asthma affects 100-150 million people worldwide. It accounts for 180,000 deaths annually. In India, it is estimated that approximately 15-20 million people are affected. In 2002, the overall prevalence of lifetime asthma in the United States was 11.9% (N = 24.7 million) 1.

Bronchial asthma is a syndrome, characterized by chronic inflammation of the airways, development of airway hyper reactivity and recurrent reversible airway obstruction.2. Clinically, asthma is expressed by airway obstruction that involves inflammation of the pulmonary airways and bronchial hyper responsiveness that is usually reversible. The past decade has witnessed phenomenal increases in the incidences of asthma, asthma-related deaths, and hospitalization.3Psychological distress, leading to depression, might be a risk factor for asthma-related morbidity and mortality4

6.2 REVIEW OF LITERATURE


  • Pistacia chinensis mainly grows in Residential scale, mainly including parts of the Central and West China where it is used as a medicine. It is used in the treatment of coughs, phthisis, asthma, dysentery etc. Resin from the closely related P. lentiscus is analgesic, antitussive, expectorant and sedative.5

  • Six compounds have been isolated from the leaves of Pistacia chinensis, a species of Anacardiaceae family. Their structures were identified on the basis of UV, IR, NMR, and MS as Gallic acid, m-digallic acid, quercetin, 6-0-galloyl arbutin-quercitrin and quercetin-3-0(6''-galloyl)-beta-D-glucosides. .6

  • Activity-guided fractionation of twigs of Pistacia chinensis resulted in the isolation and characterization of two novel ingredients as potent estrogen agonists.7

6.3 OBJECTIVE OF THE STUDY

The main objective of the proposed work is to evaluate the beneficial effects of the aqueous extract of Pistacia chinensis for its activity using various parameters in the following antiasthmatic activity models.



MATERIAL AND METHODS

7.1. Source Of The Data :

Whole work is planned to generate data from laboratory studies i.e. experiments are performed as described in references. Experimental studies in journals and in text books available with college and various institutions.



7.2 METHOD OF COLLECTION OF THE DATA:

Data collected from the different models of antiasthmatic activity done in the laboratories and journals.



METHODOLOGY :

1.BRONCHIAL HYPERREACTIVITY:8

Animals: Male Albino Guinea pigs will be procured and maintained under standard conditions with access to food and water ad libitum.

Six male Albino guinea pigs /group are used

Following groups will be made:

Group 01: Control

Group 02: Test dose 1

Group 03: Test dose 2

Group04: Standard

1. Measurement of percentage of preconvulsion time is calculated versus controls.



2. SPASMOLYTIC ACTIVITY IN ISOLATED GUINEA PIG LUNG STRIPS:8

Albino Guinea pigs of either sex are used. Each group having six Guinea pigs.

Following groups will be made :

Group 01: Control

Group 02: Test dose 1

Group 03: Test dose 2

1. Measurement of the percent inhibition of spasmogen induced contraction is calculated.



3. STABILISATION OF MAST CELLS USING RATS SENSITISED BY HORSE SERUM:9

Albino rats of either sex are used for this study. Each group having six Albino rats.

Following groups will be made :

Group 01: Horse serum + Bordetella pertusis

Group 02: 0.9% saline (without sensitization.)

Group 03: Predinisolone

Group 04: Test dose 1

Group 05: Test dose 2



parameter:

1. Measurement of percentage of degranulated and intact mast cells.


STASTICAL ANALYSIS

. The data will be analyzed using one-way analysis of variance (ANOVA).

Yes, The above study requires investigation on animals. The effect of drug will be studied on

various parameters using Guinea pigs and rat as animal model.
