PROTOCOL FOR THE INVENTORY OF V. vinifera subsp. sylvestris

WP4 : Study of Vitis sylvestris germplasm

Coordinator: J.E. Eiras-Dias

1 – Prospecting of wild grapevine populations in Europe
The inventory of V. v. sylvestris sites in Europe and the evaluation of their preservation status are very important for planning the identification and the preservation of the taxon.

With the aim of identifying wild Vitis vinifera sites (populations), each partner will characterize two sets of information :

  1. Set of descriptors applied for sylvestris population inventory, according to “European Vitis database passport descriptors” (see Work Package 61), concerning geographical location and

  2. Complementary descriptors for the evaluation of preservation status.

  1. Descriptors applied for sylvestris populations, according to “European Vitis database passport descriptors”.

For each descriptor, examples and explanations are given for sylvestris population inventory, only when the use is different than for grapevine accessions held in collection.

1. Institute code (INSTCODE)

3. Collecting number (COLLNUMB), used as Population number that is to say original number assigned by the person in charge of the population description.

Example: JED07

For the identification of wild Vitis silvestris site, we should use the initials of the responsible followed by a number with two numerals. When we describe the vines (§2), we will use the same number and we will add two numerals, which identify the individual in the population.

11. Accession name (ACCENAME), used as Population name.

13. Country of origin (ORIGCTY)

14. Location of population site (COLLSITE)

15. Latitude of population site (LATITUDE): only one value for the estimated centre of the population.

16. Longitude of population site (LONGITUDE): only one value for the estimated centre of the population.

17. Elevation of collecting site (ELEVATION): notation of a single mean value or two values for minimum and maximum elevation.

Example 1: 763

Example 2 : 245-634

29. Decoded institute (COLLDESCR)

  1. Complementary descriptors for the evaluation of preservation status

NumbPlants: number of plants observed in the population

NumbFemale: number of observed male plants

NumbMale: number of observed female plants

PresSativa: number of V. vinifera subsp. sativa near the population

Examples: 0 ; 5

PresVitis: number of other Vitis species near the population

Examples: 0 ; 3

DistanceOther: If there are other Vitis species or vinifera near the population, the distance from the nearest wild grapevine will be indicated in meters.

PopStatus: status of the population. For each site, the Partner will evaluate as very good (9), good (7), regular (5), bad (3) or very bad (1).

For a good preservation of the population the presence of a minimum number of male and female plants is needed. In this first step of GrapeGen06, each Partner will evaluate the populations using their own criteria. In the following step, when we have to consolidate the evaluations of all the populations, we will define homogeneous criteria for all the European populations. The absence of male or female plants is enough to consider the state of preservation of the population as very bad.

Example: 3

PopRisk: Finally, the Partner will indicate the risk of extinction of the population as very high (9), high (7), medium (5), low (3) or very low (1).

Example: 5

PopJustif: Justification of both status and risk of extinction of the population.

Example: Not enough female. Highway project in 2 years closed to the site.

LandOwner: Owner of the land.

Example: Private ; State ; Communal ; District

2 – Description of sylvestris plants

  • In-situ description : none (will be discussed again in Workshop 2).

  • Ex-situ description (plants collected and introduced in genebanks), starting on year 2 (2008) :

    • Passport data : the rules to follow and the single table to fill in, are the same than for WP6.

    • Primary descriptors : will be discussed again in Workshop 2.

    • SSR identification : 9 markers as decided in WP1 (will be discussed again in Workshop 2).
