BRIEF RESUME OF THE INTENDED WORK:
Need for the study :
An Antioxidant is a molecule capable of slowing or preventing oxidation of other molecules. Antioxidants are believed to quench free radicals. Free radicals are atoms or molecules with ‘singlet’, i.e. unpaired electron which makes them highly reactive. Oxidative free radicals are generated
by metabolic reactions, create a chain reaction leading to membrane and other lipid peroxidation, DNA damage etc. This has been implicated in atherosclerosis (Oxidized LDL is more atherogenic) cancers, neurodegenerative diseases and inflammatory bowel diseases. Many endogenous and dietary compounds like super oxide dismutase, ferritin, transferrin, ceruloplasmin, Tocopherol, carotene and ascorbic acid have antioxidant and free radical scavenging properties. 1
plants such as Allicin, Amla, and Turmeric are used to treat various inflammatory diseases like Cancer, atherosclerosis, premature aging, immuno-stimulant properties, as well as anti-inflammatory in which free radical oxidative stress has been implicated in the pathogenesis of these disorders.
Ayurveda and various traditional system of medicine claim that
Roxb has been used in the treatment of various diseases like delirium high fever
diarrhea ,tuberculosis and cancer and leishmaniasis. And the plant contains 4
Diospyrin , β- sitosterol, α –amyrin , β- amyrin, Lupeol , Taraxerol, Stigmasterol,
Betulin Oleanolic acid Betulinic and, Kaempferol
Studies showed that several natural products
possessing antidiabetic,antitumor and also
flavonoids containing drugs can act as antioxidant. Till now no scientific papers are
available on the leaves of
Diospyros Montana Roxb for antioxidant activity. Hence this
research work has been taken up.
6.2 Review of literature
Introduction to the Plant:
Botanical Name : Diospyros Montana Roxb.
Family : Ebenaceae
Gujrati : Timbaroa
Sanskrit: Kaaka thinduka
6 Morphology :
Diospyros Montana Roxb is a small tree with slender stem and smooth grey bark
young shoots glabrous or pubescent. Leaves alternate , ovate-oblong or elliptical acute or
sub acuminate, base usually rounded , softly pubescent or tomentose when young ultimately

Antimycobacterial activity of diospyrin derivatives and a structural analogue of diospyrin against
Mycobacterium tuberculosis in vitro
Diospyrin was isolated from the stem-bark of D. Montana Roxb The activity of diospyrin against
drug susceptible and -resistant strains of
M. tuberculosis has been reported .Three derivatives
and one structural analogue of diospyrin were
synthesized and investigated
for their inhibitory activity against Mycobacterium tuberculosis employing the rapid radiometric
method in vitro
Reported Works:- B.N. Shina et.al Reviewed the Phytochemical and Biological Studies of Diospyros Species
used in folklore medicine of Jharkhand.
N. Lall, M. Das Sarma et.al Isolated Diospyrin from the stem-bark of D. Montana Roxb and
reported the activity of diospyrin against drug susceptible resistant strains of M. tuberculosis.
Three derivatives and one structural analogue of diospyrin were synthesized and investigated
for their inhibitory activity against Mycobacterium tuberculosis employing the rapid
radiometric methodin vitro.

6.3 The objectives of the present study are:
Pharmacognostic Evaluation of Diospyros Montana Roxb . Leaves.
Extraction and phytochemical analysis.
Phytochemical investigation of various extracts of Diospyros Montana Leaves.
In- vitro evaluation of the extracts for antioxidant activity.
Isolation and characterization of phytochemical constituents from extracts having maximum activity using column chromatography and Spectral studies respectively.

Method of collection:
Collection: - The plant Diospyros Montana Roxb . will be collected from the local
Surrounding areas of Hubli, Mundagod and Katur.
Authentication: - Renowned botanist will authenticate the plant.
Pharmacognostic studies:- Pharmacognostic evaluation like morphology,
macroscopic and microscopic. Evaluation will be carried out on
Diospyros Montana leaves.
Roxb Phytochemical studies: - For the present study leaves of Diospyros Montana
will be collected shade dried and coarsely powdered .Extraction
will be done by using different solvents

extracts using DPPH free radical scavenging assay, Nitric oxide

Isolation and characterization :- The phytoconstituents from extracts having
maximum activity will be isolated and characterized by using
column- chromatography and spectral analysis respectively.
